* This slide shows how easy it is to install an Alpha unit. It literally takes seconds. We will give you all an opportunity to do this for yourselves. * Some points to note: Gradient is standard on all 96 well single blocks (inc. Moto and Chromo4) Gradient on DNA Engine line is L to R (12 different temperatures) Flat block can be used for in situ and other slide based assays, also acts as a platform for other apparatus that needs to be heated/cooled. (Has screw points for attachment.) Dual Alphas are independently programmable Moto Alphas can be used for robotic integration but also for non-robotic low volume reactions, too. * The only thing stopping the Chromo4 going on other DNA Engine line chassis is firmware; we’re working on it. * 4 x 20 character LCD display on DNA Engine and PTC-100. Press ? and ? arrows to scroll through menus (this will be easier if you have a demo cycler close by). Selected item will be in ALL CAPS. From Main Menu, select ENTER to enter a protocol. Enter name of protocol, then you have an option to add one of the following steps: Temp, Gradient, Ramp, End. When a TEMP step is selected, enter temperature and time. To include temperature/time increment/decrement, select OPTION. A GRADIENT step inserts a gradient (this is possible at any step) A RAMP step allows ramp rate to be specified Like in Basic programming, an END step is necessary to let the cycler know that a program has finished. * Reiterate that the DNA Engine line includes: DNA Engine, Dyad, Disciple, Tetrad2. All will give the same performance with the same Alpha unit. Gradient standard on 96w Alpha. * The full name is “DNA Engine Dyad”, however this is a bit of a mouthful, so we usually just use “Dyad”. * On the left, display of block/sample/lid temperatures in real time.
