CEBPα,GATA1involvedinthedifferentiationofmyeloidcells.Clinical studiesalsohaveshownthatthemutationsanddysfunctionofthetranscriptfactorsarefrequentilyexitedinhumanacutemyeloidleukemia.Previousstudiesthattheexpressionof transcriptfactorweredetectedbyRNAhybridizationduringNB4andHL-60cellsdifferentiationinducedbyATRA,theexpressionlevelsofCEBPαweredecreasedsignificantlyin NB4 and HL-60 cells, theexpression levels of GATA1were increasedinNB4cellsbyATRA,andGATA1wasnotdetectedintheHL-60cell.HoweverithavebeenreportedthattheexpressionleveloftranscriptfactorsduringNB4,HL-60cellsinducedbyAS2O3.Inthisstudy,weusedreal-timeRT-PCRtodetecttheexpressionchangesof IL-3R system(GM-CSFRα,IL-3Rα, Hβc)and transcriptfactors (PU.1,C/EBPfamily,GATA1,GATA2)duringNB4andHL-60cellsdifferentiationandapoptosisinducedbyATRA,AS2O3,andstudythattheGM-CSF、IL-3affectthedifferentiationandapoptosisofNB4cell.Results:ATRAinducedNB4celldifferentiationinthedoseof1μMaccompaniedtheexpressionsofIL-3Rα,HβcmRNAincreased.whileATRAinducedHL-60celldifferentiationinthesamedose,andaccompaniedtheexpressionsofGM-CSFRα, HβcmRNAincreased.AS2O3inducedNB4celldifferentiationandapoptosisinthedoseof1μM, andaccompaniedtheexpressionsofIL-3Rα,HβcmRNAincreased，butitisnotmoresignificantlythanNB4celldifferentiationinducedbyATRA.AS2O3cannotinduceNB4celldifferentiationandapoptosisinthesamedose,andtheexpressionsofGM-CSFRα,IL-3Rα,HβcmRNAarealsonotchanged.TheexpressionlevelsofPU.1,CEBPemRNAincreased,andtheexpressionlevelsofCEBPα,CEBPγ,GATA1,GATA2mRNAdecreasedduringNB4celldifferentiationinducedbyATRA.TheexpressionlevelsofPU.1,CEBPαincreaced,andtheexpressionlevelsofCEBPγ,GATA1,GATA2mRNAdecreasedduringHL-60celldifferentiationinducedbyATRA.TheexpressionlevelsofCEBPε, GATA1 mRNA increased, andtheexpressionlevelsofCEBPα,CEBPγ mRNA decreasedduringNB4celldifferentiationandapoptosisinducedbyAS2O3.TheexpressionlevelsofCEBPαmRNAincreased,andtheexpressionlevelsofCEBPε, CEBPγ,GATA1mRNAdecreasedinHL-60celltreatedbyAS2O3.GM-CSFandIL-3cannotstimulateNB4ce
