Study on Influence of CXCR4 Knockdown on VEGF, MMP-9, and Metastasis of Prostate Cancer CellAbstractResearch backgroundProstate cancer (PCa) was the common malignant tumor of the urinary system in old men and its metastasis occurred in early stage. It was reported that about 70% of prostate cancer presented bone metastasis in advanced stage, and the patients suffered from cancer pain, with poor quality of life and shorter survival. Moreover, as prostate cancer was not sensitive to radiotherapy and chemotherapy, radiotherapy and chemotherapy couldn’t prolong lifetime of patients. Therefore, it was very important to deeply study molecular mechanism of prostate cancer metastasis and effectively prevent the metastasis for improving the prognosis of prostate cancer.Tumor metastasis was the interacted, continuous, complex process of cancer cell and host cell, and this process was jointly completed by multiple-steps, multiple-factors interactions. Recent studies showed that chemokine CXC motif receptor4 (CXCR4) binding to its ligand stromal cell-derived factor-1 (SDF-1) played an important role in tumor angiogenesis and invasion metastasis. Our research group previously found that CXCR4 expression was positively associated with microvessel density and lymph node metastasis of patients with prostate cancer, indicating the receptor could promote prostate cancer angiogenesis and metastasis. However, action mechanism and therapeutics significance of CXCR4 promoting prostate cancer metastasis should be still further investigated.Recently, effects of vascular endothelial growth factor (VEGF) and matrix metalloproteinases-9 (MMP-9) in tumor have been increasingly regarded. High expressions of VEGF and MMP-9 in various kinds of tumors are regarded as important impact factors of tumorigenesis, invasion metastasis, treatment resistance and recurrence.
