维普资讯 解剖学研究 2008年第 30卷第 3期 AnatRes。2008．Vo1．30．No．3 · 论 著 · 传代培养 PC12细胞的分泌特性及 PENK基因表达的研究 马 超 伍少玲 燕铁斌 (中山大学附属第二医院康复医学科 ，广东 广州 510120) 摘【要】 目的 观察Pc12细胞传代培养中的生长情况，检测培养液中去 甲肾上腺素(NE)和甲硫氨酸脑啡肽(M． EK)的分泌水平及细胞前脑啡肽 (PENK)基因的表达情况，为细胞移植镇痛的研究提供实验依据 。方法 PC12细胞在 DMEM+15％胎牛血清中培养 ，酶法消化传代 ；采用高效液相色谱．电化学检测法检测培养液 中NE的浓度 ：采用放射免疫 法检测培养液中M．EK的浓度；采用反转录 PCR检测细胞 PENKmRNA的表达。结果 传代培养PC12细胞的基础分泌 ： NE为 (465．8+78．5)pg／mL，M．EK为 (4．3+0．66)pg／mL；PENK低水平表达。结论 PC12细胞在传代培养过程中，可合成和 分泌 NE和M．EK，并可检测到PENK基因的表达 ，为细胞移植镇痛提供新的细胞来源。 【关键词】 PC12细胞； 脑啡肽； 前脑啡肽基因 Characteristicsofsecretion and geneexpression ofPENK in serialsubcultivated PC12cells MA Chao， Shao-ling， YAN Tie-bin．DepartmentofRehabilitationMedicine， TheSecondAffiliatedHospital， SunYat-senUniversity， Guangzhou 51o120China A【bstract】 Objective ToinvestigatethegrowthcharacteristicsofPC12cellsanddetectthelevelofsecretionandthe expression ofthe proenkephalin mRNA in serialsubcuhivation， and to provide the evidence ofcelltransplantation in pain treatment．Methods PC12cellsweregrowninDMEM supplementedwith15％fetalbovineserum．Thenorepinephrine(NE)in themediumwasmeasuredwithhigh-performanceliquidchromatography-electrochemicaldetectiontechnique(HPLC-ED)，and methionine-enkephMin(M-EK)wasmeasuredwithRadioimmunoassay．Theproenkephalin(PENK)mRNA expressioninPC12 cellsweredetectedbyreversetranscription-PCR． Results ThelevelsofbasicsecretionofNE andM-EK inthemedium were (465．8+785)pg／mLand(4．3±0．66)pg／mLrespectively．ThePENKmRNA inPC12cellswerelowexpression．Conclusion Th ePC12cellsculturedinvitrocouldsynthesisandsecreteNE and M—EK，andtheexpression ofPENK mRNA waspositive．It willprovideanew cellresourceofcelltransplantation inpaintreatment．
