汕头汕头大学医学院硕士学位论文 Ⅲ Ⅲ Abstract Objective To investigate the correlation of IL-7 level with T lymphocyte subsets and CD4+CD25+Treg in patients with chronic hepatitis (CHB). Methods 1Electing 35 chronically HBV-infected patients,14 Hepatitis B carriers (AsC),18 patients(received antiviral therapy，Rlt) and 14 normal controls(NC).Detecting the percentage of T lymphocyte subsets and CD4+CD25+Treg in the peripheral blood of those groups with flow cytomerty; The expression of IL-7 in the supernatants were measured by ELISA; Measuring the serum of HBV-DNA in four groups with real-time RT-PCR; Measuring the serum of ALT in all of the groups with AU5400 biochemistry analyzer; Results 1 The expression of IL-7 of patients with CHB was lowest in the three groups (F=437，P0.05), the group (Rlt) was lower than the CHB (P0.05). The percentage of CD4+T in four groups were（33±2.9）%、（38.64±6.78）%、（43.2±9.4） %，（39.8±3.1）%, and CD8+T were （23.6±4.71）%、（27.5±3.31）%、（30.5±8.6）%，（32.46±3.81）%.The CD4+CD25+Treg from group CHB was extremely high when compared with the other two groups,the percentage in group CHB,AsC and NC were respectively（9.82± 1.12）%、（7.29±0.95）%、（4.19±1.11）%，(F=138.6. P0.05). The significant difference found in the expression of Treg 、CD4+T 、CD8+T between the CHB group and the group（Rlt）(F=51， P0.05，F=28，P0.05，F=25，P0.05). 3 In CHB,IL-7 showed significant correlations with CD4+CD25+Treg (r=-0.381 ， P0.05) , CD4+T（r=-0.530，P0.05）and CD8+T（r=0.496，P0.05）.Furthermore，the expression levels of IL-7 from group CHB were negatively associated with HBV-DNA（r=-0.494，P0.05）,but not with ALT （r=0.317，P0.05）. Ⅳ Ⅳ Conclusion There exists immunologic dysfunction in the patients with chronic hepatitis B,make the activation of immune cell and IL-7 Abnormally. Nucleoside drugs can effectively inhibit HBV replication,reduce the bodys abnormal immune activation levels to a certain extent,which still cant be recovered completely.During antiviral process,IL-7 level changes gruadually,suggestin
