万方数据 万方数据 摘要 脂蛋白脂酶（Lipoprotein lipase，LPL）基因是一种在脂肪代谢过程中起关键作 用的蛋白水解酶。LPL 基因已被看做是影响 IMF 的候选基因。提高山羊脂肪沉积能 力，有利于获得与山羊肌肉脂肪性状相关的标记位点。本研究选用安徽白山羊、波 尔山羊、马头山羊、济宁青山羊、岷山黑裘皮羊、临朐奶山羊等 6 种山羊群体为试 验动物，采用 PCR-SSCP 分析方法研究 LPL 基因的多态型，并运用 GLM 模型，对 检测出来的的 SNP 及其与羊 IMF 的关联分析。 1. 根据 GenBank 上公开发表的相关山羊 LPL 基因，设计引物 P1-P10，应用 PCR- SSCP 分析方法在 LPL 基因上发现 P1-SSCP 多态位点，该位点上具有五种基因型 AA、 BB、AB、AC 和 BC。序列测定结果表明，P1-SSCP 的变异是由碱基 G→287A 的替 换造成的。在 P6-SSCP 多态位点上发现三种基因型 AA、BB、AB。序列测定结果表 明，P6-SSCP 变异是由碱基 G→16997A 的替换造成，Gln→Ser。在 P7-SSCP 多态位 点上发现三种基因型 AA、BB、AB。序列测定结果表明，P7-SSCP 变异是由碱基 A →18313C 替换造成，导致 Asp→Ala。 2. 在上述 6 个群体中分析了 PCR-SSCP 位点的基因型分布情况，Hardy-Weinberg 检验结果表明：在 P1-SSCP 位点上，除安徽白山羊之外，所有品种均达到 Hardy- Weinberg 平衡（P0.05）。在 P6-SSCP 位点和在 P7-SSCP 位点上，所有品种均达到 Hardy-Weinberg 平衡状态（P0.05）。 3. 根据 SNP 群体动力学分析，287G、18313A 在 6 个群体中分布均较稳定， 16997G 在群体中表现一定特异性，安徽白山羊分布达 0.9750，波尔山羊 0.8828，马 头山羊 0.8333，临朐奶山羊 0.7500，济宁青山羊 0.7037，岷山黑裘皮山羊中分布率 最低是 0.6979。G-16997A 与 IMF 呈负相关性；A-18313C 位点与 IMF 显著相关联 （P0.05），这说明 P7 位点处 A-18313C 与脂肪沉积能力有相关性。 关键词：山羊；LPL 基因；遗传多态性；肌内脂肪含量 I Abstract Lipoprotein lipase (LPL) plays a major role in the metabolism and transport of lipids. LPL gene has been seen as a candidate gene affecting intramuscular fat content. In order to obtain marker loci about goat muscle fat traits and improve the ability of fat deposition, six goat breeds selected were Anhui white goats , Boer goats, horse goat, Jining Grey goats , minshan black fur sheep, and LinQu goats. The six populations were regarded as experimental subjucts. The polymorphisms of LPL were examined using PCR-SSCP, analyzed by using GLM model and to analyse the relationship between the polymorphisms and intramuscular fat content in goats. The specific primers p1-p10 were designed according to the relevant goat LPL gene sequences published in GenBank. The P1 polymorphic loci has been found in the LPLgene using PCR-SSCP, with five genotypes AA、BB、AB、AC and BC on the site. The measurement results show that the P1 variation is replaced by the nucleotide G→287A.
