第38卷第8期 南京医科大学学报（自然科学版） 2018年8月 ACTAUNIVERSITATISMEDICINALISNANJING（NaturalScience） ·1039· 糖尿病小鼠胰腺组织Netrin⁃1表达及意义 李 琪，谷庆炜，曹 欣，李 倩* 南京医科大学附属南京医院内分泌科，江苏 南京 210006 ［摘 要］ 目的：研究糖尿病小鼠胰腺组织中Netrin⁃1的表达并探讨其在糖尿病发生过程中的作用。方法：C57BL/6J小鼠随 机分为正常对照组和糖尿病模型组，应用RT⁃PCR和Westernblot方法分别检测小鼠胰腺组织中Netrin⁃1mRNA和蛋白的变化； 在正常和异常葡萄糖浓度下培养大鼠胰岛细胞瘤细胞（ins⁃1），MTT检测细胞活性，RT⁃PCR和Westernblot方法分别测定Netrin⁃1 mRNA和蛋白的表达情况。结果：糖尿病小鼠胰腺组织内Netrin⁃1的mRNA和蛋白表达增加，高浓度葡萄糖刺激ins⁃1细胞上 调Netrin⁃1mRNA和蛋白的表达。结论：糖尿病小鼠胰腺组织表达Netrin⁃1的水平增加，这与血糖异常相关，Netrin⁃1可能与糖 尿病的发生发展相关。 ［关键词］ 2型糖尿病；胰腺；Netrin⁃1；血糖；ins⁃1细胞 ［中图分类号］ R587.1 ［文献标志码］ A ［文章编号］ 1007⁃4368（2018）08⁃1039⁃04 doi：10.7655/NYDXBN ExpressionandsignificanceofNetrin⁃1inpancreatictissueindiabeticmice LiQi，GuQingwei，CaoXin，LiQian* DepartmentofEndocrinology，NanjingFirstHospital，NMU，Nanjing210006，China ［Abstract］ Objective： TostudytheexpressionofNetrin⁃1inpancreatictissueindiabeticmiceandexplorethesignificanceinthe developmentoftype2diabetesmellitus. RT⁃PCRand WesternblotwereusedtodetectthemRNAandproteinofNetrin⁃1respectively.Results：ThemRNAandproteinexpressionofNetrin⁃1 increasedinpancreatictissueofdiabeticmice.Whilehighconcentrationofglucoseup⁃regulatedthemRNAandproteinexpressionof Netrin⁃1inins⁃1cells.
