The sources of stem cell, the means of cell delivery, and the restoration of structure and function of the optic nerve were discussed. Based on the current research, the potential benefit and risk of stem cell therapy for the glaucoma patients was evaluated. Some suggestions were given for the future research on theses issues. (Ophthalmol CHN, 2009, 18: 10-13) 【Key words 】glaucoma; stem cell therapy 青光眼是全球第一位不可逆致盲性眼病。通过药物或手术降低眼压，以阻止进一步的 损害一直是临床唯一的治疗方法。虽然有关神经保护的研究已经持续多年，但至今仍缺少肯 [1] [2] 定的结果。其他可能有效的治疗主要为抗原接种诱导保护性自体免疫 和增加眼部血流 。 [3] 最近十年，干细胞的突破性研究，例如，2008 年美国《科学》杂志报告 ，从肌萎缩性(脊 髓) 侧索硬化症（ALS ）患者体内提取的成熟细胞，经基因编程转为具有增殖和分化成运动 神经元的潜能，使临床医师考虑到细胞替代疗法治疗神经变性疾病（包括青光眼）的可能性。 所谓细胞替代疗法是指用正常的细胞替代死亡的细胞以恢复原有的机体功能的方法。青光眼 作为视网膜神经节细胞进行性凋亡（或死亡）性疾病，在理论上存在着应用细胞替代疗法尤 其是干细胞治疗的可能性。 干细胞（stem cell ）是具有自我更新、高度增殖和多向分化潜能的细胞群体。它既存在 于胚胎发育阶段，如胚胎干细胞（embryonic stem cells ，ESC ）及某些器官的前体细胞 （progenitor cells ），也存在于发育成熟的个体的某些部位，如造血干细胞、骨髓间充质干 细胞（bone marrow mesenchymal stem cells ，MSC ）、角膜缘干细胞和视网膜干细胞等。利 用干细胞为种子细胞，生成新的组织，对病变组织进行替代、修复和功能重建，一直是研究 [4-5] 的热点。在眼部，很早就开始了干细胞在
