毕业设计(论文) 课 题 名 称 2-苯并噁唑肼对葛根中酪氨酸酶 抑制作用的研究 2-苯并噁唑肼对葛根中酪氨酸酶抑制作用的研究 摘 要 本文旨在通过单因素实验探究新型抑制剂2-苯并噁唑肼（HBO）对葛根中酪氨酸酶的抑制效果。然后通过三因素三水平正交实验，以抑制剂的浓度、作用时间、作用温度等作为考察因素，以对底物的抑制率作为指标，通过SPSS软件进行数据处理，得到最佳的反应条件。在单因素试验试验中最佳抑制剂浓度为40μmol/L、最佳作用时间为430s、最佳作用温度为35℃。最佳反应条件是在葛根与磷酸盐缓冲液（PBS）的配比为0.5g:2ml、抑制剂浓度为35μmol/L、作用时间为370s、作用温度为30℃时，可达到对葛根中酪氨酸酶的最佳抑制效果。最终的结果显示，HBO对葛根中酪氨酸酶有较好的抑制效果。通过本课题的研究，为高效酪氨酸酶抑制剂的开发及应用奠定了基础。 关键词：葛根；酪氨酸酶；2-苯并噁唑肼；正交试验；抑制效果 Studying on Inhibition effect of 2-benzoxazolyl hydrazine on the Pueraria  Xu Ying(College of Biological and Chemical Engineering, Shaoyang Shaoyang ,Hunan Abstract This thesis aimed to explore the new single factor inhibitor 2-benzoxazolyl hydrazine (HBO) of puerarin in the inhibitory effect of tyrosinase. By three factors and three levels of orthogonal experiment，taking inhibitor concentration, reaction time, reaction temperature as investigation factors and the substrate inhibition as indicator, throught SPSS for data processing, the best reaction conditions can be obtained. In the single-factor test, the optimum inhibitor concentration was 40μmol/L, the optimum action time was 430s, and the optimum reaction temperature was 35℃. When the optimum conditions were pueraria and phosphate buffer solution (PBS) in the ratio of 0.5g: 2ml, the inhibitor concentration of 35μmol/L, reaction time was 370s, the role of temperature was 30℃，pueraria can achieve the best inhibitory effect in tyrosinase. The final results showed that HBO had a good tyrosinase inhibitory effect on pueraria. Through research of this subject, a good foundation was laid for efficient tyrosinase inhibitor development and application.
