重庆大学硕士学位论文英文摘要 重庆大学硕士学位论文 英文摘要 II II ABSTRACT SCF coplex/26S proteasome pathway is a major mechanism underlying many organismal processes, such as hormone signal transduction, cell division, flower development, stamen recognition and stress response. As is one of components of SCF complex involved in ubiquitin proteasome system, F-box protein combines with SCF complex through F-box domain in N termini, and determines target proteins with different motifs in C termini. This study separates a novel F-box gene of tomato, named SlFbf, involved in stamen and fruit development. We analyze expression mode of SlFbf and hormonal signaling SlFbf probably involved in, additionally reveal functions of SlFbf with transgenic method. The main conclusions are as follows: We analyzed construction features and expression mode of SlFbf through prediction tools of bioinformatics. There are four characters about SlFbf: firstly, SlFbf contains complete ORF coding 381 amino acids with F-box motif in N termini and FBA_1 motif in C termini; secondly, SlFbf are intron-free in tomato genome; thirdly, fragments of SlFbf exits in leaf, fruit and callus; finally, predicted promoter of SlFbf includes GA, ABA and lots of light response elements. The expression mode showed that SlFbf has weak expression in root and green fruit, moderate expression in stem, leaf, alabastrum, yellow and red fruit, the strongest expression in flower. SlFbf exhibits remarkable strong expression in stamen at anthesis compared with pistil, petal and sepal during other three periods including -2 dpa, 4 dpa and 8dpa. The different expression characters of SlFbf in different tissues demonstrated that SlFbf functioned in stamen and fruit development. Treating seedlings with exogenous hormones including ABA, GA, ETH and NAA, we found that SlFbf was up-regulated by GA, down-regulated by ABA, but had no obvious response to NAA and ETH.
