The use of mouse embryonic fibroblasts (MEF) as feeder layer exists the problem of pollution of animal pathogens. In this paper, the characteristics of human amniotic epithelial cells (HAEC) and its use as a feeder layer of embryonic stem cells are described. The genomic DNA methylation in embryonic stem cell differentiation is also explored. Optimization and standardization of culture methods is needed for research as well as for clinical purpose.
