国际中医中药杂志2019年3月第41卷第3期 Int J Trad Chin Med, March 2019, Vol. 41, No.3  ·279 · ·实验与方药 · RP-HPLC 法同时测定三黄片中 4 种有效成分含量 1 2 邹沫君 任磊 1 2 四川省乐山市食品药品检验检测中心食品检测室 614000 ； 乐山师范学院化学系 614000 通信作者：邹沫君，Email: 305898151@，电话 【摘要】 目的 建立同时测定三黄片中盐酸小檗碱、黄芩苷、大黄素、大黄酚含量的 RP-HPLC 法。 方法 采用 Thermo Hypersil GOLD aQ 色谱柱(250 mm ×4.6 mm ，5.0 μm)，以甲醇-0.1%磷酸水溶液为流 动相，进行梯度洗脱，流速 1.0 ml/min，柱温30 ℃，检测波长254 nm 。结果 盐酸小檗碱、黄芩苷、大黄素、 大黄酚的分离度均符合要求，线性范围分别为 0～0.634 8 μg (r ＝0.999 9)、0～1.017 2 μg (r ＝0.999 9)、0～0.802 4 μg (r ＝0.999 9)、0～0.967 2 μg (r ＝0.999 9)；平均回收率(n ＝9)分别为 101.03%、99.81%、99.35%、 99.54% ；定量限分别为0.036 3、0.021 0、0.049 7、0.079 3 mg/g ；供试品溶液在24 h 内稳定，RSD (n ＝5) 分别为 0.48%、0.56%、0.68%、0.49%；方法耐用性良好，在不同柱温(±1 ℃)条件下，RSD (n ＝3)分别为 0.57%、0.42%、0.44%、0.65%，不同流速(±0.1 ml/min)条件下，RSD (n ＝3)分别为 0.74%、0.29%、0.46%、 0.56%。12 批样品中盐酸小檗碱、黄芩苷、大黄素、大黄酚含量分别为 4.630 3～5.586 6、14.551 4～18.818 9、 0.171 1～2.798 4 、0.529 9～2.925 4 mg/小片。结论 本法经方法学验证，适用于三黄片的质量控制。 【关键词】 色谱法，反相；三黄片；小檗碱；黄芩苷；大黄素；大黄酚；质量控制；含量测定(中药) DOI: 10.3760/cma.j.issn.1673-4246.2019.03.015 Simultaneous determination of four components in Sanhuang tablets by RP-HPLC 1 2 Zou Mojun , Ren Lei 1Leshan Center for Food and Drug Control, Leshan 614000, China; 2Department of Chemistry, Leshan Normal University, Leshan 614000, China Corresponding author: Zou Mojun, Email: 305898151@, Tel: 【Abstract 】 Objective To establish a RP-HPLC method for simultaneously determining berberine hydrochloride, baicalin, emodin and chrysophanol in Sanhuang tablets.
