國立宜蘭大學食品科學研究所 碩士班專題討論 演講者:潘雅琪 Pan, Ya-chi 題目:天然有機物抑制酪胺酸酶活性及其作用機制 指導教授:陳翠瑤 老師 Study of the tyrosinase activity inhibition and 日期:May 16, 2008 mechanism from natural organism. 中文摘要 黑色素沉澱(皮膚出現黑斑、雀斑、老人斑)會影響個人觀瞻,而酪胺酸酶為黑色素形成之主要關鍵酵素。酪胺酸酶催化酪胺酸氧化成多巴(dopa),再將多巴再轉換成多巴醌後,隨即進行一連串反應而形成黑色素。酪胺酸酶抑制劑在美白化妝品中扮演重要角色,採用天然有機物抑制酪胺酶預防黑色素沉澱之優點為具有豐富的生物活性及無危害人體的顧慮。天然有機物抑制酪胺酸酶活性結果顯示:肉桂酸衍生物―3,4-dihydroxycinnamic acid及4-hydroxy-3-methoxycinnamic acid分離自翁草根部,抑制酪胺酸酶活性之IC50分別為0.97 mM 及0.33 mM ;類黃酮―glabrene及isoliquiritigenin分離自甘草根部,抑制酪胺酸酶活性之IC50分別為3.5 ?M 及8.1 ?M;原兒茶酸甲基酯化合物分離自黑米糠,抑制酪胺酸酶活性之ID50為0.28 ?mol/mL;縮合單寧分離自紅麴,抑制酪胺酸酶活性之ID50為5.57 mg/mL;呋喃醛衍生物―5-(hydroxymethyl)-2-furfura分離自竹笙,抑制酪胺酸酶活性之IC50為 關鍵字:酪胺酸酶、酪胺酸酶抑制劑、美白劑、黑色素 Abstract Melanin pigmentation (melasma, freckles, senile elntigines etc.) is an aesthetic problem in human beings. Tyrosinase is the key enzyme to form melanin. Tyrosinase converts tyrosine to dihydroxyphenylalanine (DOPA) and then to dopaquinone. Subsequently, it takes a series of reactions, and finally to form melanin. Tyrosinase inhibitors play an important role in whitening cosmetic. The advantages of inhibiting tyrosinase from natural organism have rich bioactive chemicals and free from harmful effects. The result of inhibiting tyrosinase activity exhibited as follows. Cinnamic acid derivatives, 3,4-dihydroxycinnamic acid and 4-hydroxy-3-methoxycinnamic acid, were isolated from Pulsatilla cernua roots, and the IC50 values were 0.97 mM and 0.33 mM, respectively. Flavonoid, glabrene and isoliquiritigenin, were isolated from licorice roots, and the IC50 values were 3.5 ?M and 8.1 ?M, respectively. Protocatechuic acid methyl ester were isolated from black rice bran, and the ID50 values was 0.28 ?mol/mL. Condensed tannins were isolated from red koji extracts, and the ID50 was 5.57 mg/mL. Furfural derivatives, 5-(hydroxymethyl)-2-furfura, was isolated from Dictyophora indusiata, and the IC50 was 0.98 mM. Inhibitor ingredients of the above, except the condensed tannin, glabrene an
