兰州大学硕士学位论文 兰州大学硕士学位论文 黄苠的糙皮侧耳发酵及其发酵液的药效研究 中文摘要 黄芪(RadixAstragali)是一种常用中药，素以“补气诸药之最著称，在中药中具有 举足轻重的地位。黄芪具有多种功效，补气开阳、益胃固表、托毒生肌、利水退肿是中医 对它的概括描述。近年来，随着黄芪用量和用途的逐年增加，沿用多年的黄芪使用方法以 及加工方法及其有效成分提取方法已不能满足社会的需求。 本论文在继承中药炮制学发酵法的基础上，结合现代生物工程的发酵技术，利用糙皮 侧耳(Pleurotus ostreatus)对黄芪进行发酵处理，以增加黄芪的疗效、提高黄芪的实际利 用率，其主要研究内容和结果如下： 利用单因素平板生长实验和摇瓶试发酵，优化黄芪发酵培养基，初步得到较佳发酵培 养基成分组成为：黄芪粉末(60目)509、麸皮(煮汁)209、石膏(60目)1卧蒸馏水 1000ml，pH值自然。 通过对黄芪发酵过程(第1、3、5、7、9、13、15d)发酵液中主要活性成分含量检测， 并与黄芪水煎液相应活性成分对比发现，发酵处理使黄芪多糖和游离氨基酸含量降低，而 总黄酮、总皂苷和黄芪甲苷含量增加，但增加幅度不高。并确定了黄芪发酵的最佳时间为 7天。 对黄芪发酵液进行药理实验研究发现：黄芪发酵液的大、中剂量对小鼠具有显著的抗 缺氧、抗凝血、利尿作用，与戊巴比妥钠合用大剂量具有延长小鼠睡眠潜伏期和缩短小鼠 睡眠持续时间的作用。但与黄芪水煎液相比，只有抗凝血活性具有显著差异性(P0．05)。 关键词：黄芪；糙皮侧耳；发酵；含量检测；抗凝血 第1I页 兰州人学硕．1：学位论文 兰州人学硕．1：学位论文 黄芪的糙皮侧耳发酵及其发酵液的药效研究 Abstract Radix Astragali is one commonly used traditional Chinese medicine，usually being known as”the best of all medicine of tonifying Qi”．It has decisive position in traditional Chinese medicine and has many curative effects．In recent years．wim the increase of Radix Astragali consumption,the traditional methods of Radix Astragali usage and processing and active components extraction have been unable to meet the needs of the community． In combination with the modem bio-engineering fermentation technology on the basis of the zymotechnics of traditional Chinese medicine processing，we utilized Pleurotus ostreatus to ferment Radix Astragali．The objection of our work is to achieve its high efficacy and the actual utilization．The results showed that： Using the single—factor of plate growth experiment and liquid flask shaking experiment，the optimal composition of the cultrue medium for Radix Astragali is：Radix Astragali(60 eyes)509， bran(cookjuice)209，gypsum(60 eyes)lg，distilled water 1000ml，pH value Was natural． The major active components were detected in the process(the first,third，fifth,seventh， ninth，eleventh，thirteenth，fifteenth day)of Radix Astragali fermentation．Compared、)l，ith the decoction of Radix Astragali，the contents of soluble polysaccharids an
