Cance r I ns ti t ute , Chi nese A ca demy of M e d ical S cience Abstract :Objective 　To ob serve whet her in sulin can induce an enhancement of chemot herap eutic respon se of 5fluorouracil (5Fu) on human t ran splant able esop hageal cancer nude model and st udy t he mechani sm of action . Methods 　The exp ression of cyclin D1 in t umor ti ssue was t est ed by immunohi stochemi st ry . Murine serum in sulinlike growt h factor 1 ( I GF1) and in sulinlike growt h factor binding p rot ein3 ( I GFB P 3) were det ermined by EL ISA . Results 　High , medium and low do se in sulin (0 . 09 ,0 . 06 ,0 . 03 u/ 20 g) combined wit h 5Fu could inhibit t he growt h of t umor greatly ( P 0 . 05) but wit hout severe side effect s of 5Fu . Comp ared wit h cont rol group , t he exp ression of cyclin D1 increased significantly in in sulin group ( P 0 . 05) . There wa s not a st ati stical significance on t he level s of serum I GF1 and I GFB P3 between combination group s and 5Fu alone group ( P 0 . 05) . Conclusion 　In sulin can enhance t he chemot hera p eutic respon se of 5Fu on human t ran splant able esop hageal cancer nude model . The increa sed exp ression of cyclin D1 play s an import ant role in t he enhancement . The change of level s in serum I GF1 and I GFB P 3 fail s to elucid
