法测定苏丹红 I 的新方法。20 ℃时,苏丹红 I 的含量在0.53~16.00µg/ml 浓度 范围内,苏丹红 I- 甲醇体系的RLS 强度降低值(ΔI )与苏丹红 I 的浓度(C 苏 丹红 I )有良好的线性关系,线性回归方程为 ΔI =16.55C 苏丹红 I +16.04,r=0.996, 方法检出限 0.16µg/ml,测定范围 0.53~16.00µg/ml, RSD =1.82%~2.41 % (m=6 ,n =11),加标回收率为 92.67%~103.80% 。方法精密度好,准确度高, 十分简单,易于推广,应用于实际样品的检测,结果满意。 第四章建立了反相高效液相色谱法测定痕量苏丹红 I 的新方法。以四氢 呋喃-酸性水溶液为流动相,紫外检测器测定。在 0.20~100.00µg/ml 浓度范围 内色谱峰面积与苏丹红 I 的浓度有良好的线性关系,回归方程为 A=5.7414C +0.565,相关系数r =0.9997,检出限为 82ng/ml ,测定下限为 0.20µg/ml,RSD =0.48 %~4.34 %(m=6 ,n =11),加标回收率分别 98.07 %~99.85 %。应用于 辣椒粉样品中苏丹红 I 的检测,结果准确。 关键词:苏丹红 I;人血清白蛋白;荧光猝灭;共振光散射;高效液相色谱 2 Novel detection methods of sudan I and their applications ABSTRACT Sudan I is a genotoxic carcinogen and its presence is not permitted in foodstuffs for any purpose at any level. In this context, the rationale is on the basis that this colourant could be found in some food products, because of its intense red orange colour. For this reason, sensitive, selective and accurate analytical methods should be developed in order to identify and quantify such substance. As reported in the literature, dealing with the analytical chemistry of Sudan I, several methods were developed to detect the presence of this compound, however, some existing methods of equipment demanding. In this work, we studied the mechanism of the sudan I with HSA, proposed three novel methods to concentrate, purify and detect this substance. In the chapter 1, a novel detection has been developed for Sudan I based on the fluorescence quenching effect . The fluorescent quenching value of HSA at 337nm was linear to the concentration of Sudan I in the range of 0.47~7.50µg/ml, at 25℃, in the Tris-HCl buffer solution (pH8.2) with the detection limit was 0.14µg/ml.
