Methods Colorectal cancer SW480 cells was treated with fluorescent labeled PEG-liposomes, PEG-liposomal L-oHP, respectively. Cellular uptake of liposomes were detected by flow cyometry (FCM) and fluorscence microscopy. The cells proliferation was analyzed by MTT; Cellular ultrastructure observed through transmission electron microscopy (TEM); Apoptosis were detected by FCM and Tunel; Analysis of actived-caspase 3 (P17) expression with Western blot. Result Cellular uptake of liposomes, at 24 h, the average fluorescence intensity was 3.25 fold at 2 h. In the same dose, the PEG-liposomal L-oHP induced a strong apoptotic response than free L-oHP. The levels of apoptosis was increased, then protein expression of P17 was up-regulated. Conclusion PEG-liposomal L-oHP were reinforced induced apoptosis of human colorectal cancer SW480 cells.
