摘 要 目的：筛选超声法提取葛根总黄酮的最佳工艺条件。 方法：采用均匀设计法考察了乙醇浓度、乙醇用量、提取时间、提取温度、超声提取次数等五个因素对葛根总黄酮提取的影响；利用紫外-可见分光光度法测量葛根总黄酮的含量；以葛根总黄酮最终得率为指标进行工艺条件的优化， 结果：结果表明，最佳工艺条件为乙醇浓度75%，乙醇用量20 ml/g，提取时间40 min，提取温度68 ℃，超声5次。该提取工艺省时、节能、操作简便、得率高。 结论：影响超声辅助提取葛根总黄酮的主要因素有乙醇浓度、乙醇用量、提取温度、提取时间和超声处理次数，各因素对葛根总黄酮得率的影响强弱顺序为：乙醇浓度＞提取温度＞乙醇用量＞超声处理次数＞提取时间。 关键词：超声法；提取；葛根；总黄酮；工艺条件；均匀设计 Abstract Objective: Optimize the extraction condition of total falconoids from Radix Puerariae Lobatae by ultrasonic wave， Methods: Choosing the uniform design as method, the experiment was carried out to study the influence of alcohol content, quantity of solvent, extraction time, extraction temperature and operation times on the result of extraction. The content of total falconoids of Radix Puerariae Lobatae was determined by ultraviolet-visible spectrophotometry. The optimal extraction condition was selected with the extraction yield of total Radix Puerariae flavonoids as marker. Results: The results indicated that the optimum process was: extracting with 75% alcohol, the quantity of solvent 20 ml/g, extraction time 40 min, temperature at 68 ℃, 5 operation times. Compared with the traditional extraction methods, this extraction process is time- and energy saving, and also convenient and efficient. Conclusion: The influence of alcohol content, quantity of solvent, extraction time, extraction temperature and operation times is the primary factor on the extraction of total flavonoids from radix Pueraria Lobata Ohwi by ultrasonic wave.
