???? 健康人口服羟乙膦酸钠胶囊剂的相对生物利用度试验 ???? ???? 摘　要：目的：研究羟乙膦酸钠胶囊在正常人体内的相对生物利用度。方法：采用钼蓝-乙酸丁酯萃取比色法测定8名志愿者单剂量po 400 mg羟乙膦酸钠胶囊和羟乙膦酸钠片剂后的血药浓度变化情况，计算二者的相对生物利用度。用双单侧t检验法分析两种制剂的生物等效性。结果：两种制剂的消除相半衰期分别为(3.03±1.07)h和(3.46±0.73)h；血药浓度曲线下面积分别为(8.18±1.99)和(8.14±1.94)μg．h．ml-1，其达峰浓度分别为(0.88±0.17)和(1.04±0.32)h，达峰浓度分别为(2.05±0.56)和(1.79±0.60)μg．ml-1。结论：羟乙膦酸钠胶囊与羟乙膦酸钠片剂为生物等效制剂，羟乙膦酸钠胶囊的相对生物利用度为(100.6±10.0)%。关键词：羟乙膦酸钠；生物利用度 Relative bioavailability of disodium etidronate capsules in healthy volunteers Shao Qing (Shao Q)　Li Shimin (Li SM)　Zeng Su (Zeng S)(School of Pharmacy,Zhejiang University,Hangzhou 310031) ABSTRACT：OBJECTIVE：To study the relative bioavailability of disodium etidronate capsules in healthy volunteers.METHODS：The disodium etidronate concentrations in plasma were determined by extraction of molybdate blue-butylacetate and colorimetry following a single oral dose of disodium etidronate capsules and disodium etidronate tablets respectively given to 8 volunteers.RESULTS：The t1/2Ke was (3.03±1.07)h and (3.46±0.73)h,AUC (8.18±1.99) and (8.14±1.94)μg．h．ml-1，tmax (0.88±0.17) and (1.04±0.32)h，cmax(2.05±0.56) and (1.79±0.60)μg．ml-1 of disodium etidronate capsules and disodium etidroante tablets,respectively.CONCLUSION：The results of statistical analysis showed that the two formulations were bioequivalent.The relative bioavailability of disodium etidronate capsules was (100.6±10.0)%.KEY WORDS：disodium etidronate,bioavailability▲ 　　羟乙膦酸钠(disodium etidronate)是一新型的防治骨质疏松症的药物。目前，国外报道的血药浓度测试方法多采用同位素标记法［1～2］,我们参考S.Bisaz报道的方法［2］，经改进去除了同位素标记步骤，并采用钼蓝-乙酸丁酯萃取比色法［3～4］测定8名健康志愿者口服羟乙膦酸钠胶囊剂和片剂的血药浓度，并计算出有关的药动学参数。同时根据测得的AUC经统计分析，以评价这两种制剂的生物等效性，为该药临床应用的有效性和安全性提供依据。 1　材料与方法 1.1　药品与试剂　　试验药品：羟乙膦酸钠胶囊剂(200 mg/粒，浙江海门制药厂提供，批号960724)；对照药品羟乙膦酸钠片剂(200 mg.片-1，成都化学制药厂生产，批号960501)；羟乙膦酸钠标准品(浙江海门制药厂，含量99.52%)。　　钼锑贮存液：取分析纯浓硫酸90.5 ml，缓缓加入约200 ml水中，搅匀，冷却。另称取钼酸铵25.0 g溶于上述硫酸液中，再加0.45%酒石酸锑钾液100 ml，冷却后用水稀释至500 ml，摇匀，贮于棕色瓶中，冰箱保存。　　钼锑抗混合显色液：准确量取钼锑贮存液100 ml，加入抗坏血酸0.50 g溶解即可。该显色液保存时间为24 h，宜用前配制。　　实验中所用试剂均为分析纯。1.2　仪器　　日立200
