第9 卷第11期 Vol.9 No.11 2016 年 6 月 June 2016 CIP2A 抑制非小细胞肺癌细胞凋亡 董千泽，付 琳，韩 勇，刑济麟* （中国医科大学基础医学院，沈阳 110001 ） 摘要：目的：探讨蛋白磷酸酶2A 的癌性抑制因子（cancerous inhibitor of protein phosphatase 2A ，CIP2A ）是 否上调细胞凋亡蛋白Bcl-2 和Bcl-xl 的表达。方法：选取HBE 细胞系及A549 细胞系进行实验，通过流式细胞 术、实时PCR （real-time PCR ，RT-PCR ）和Western blotting 方法检测细胞凋亡情况及凋亡相关蛋白Bcl-2 、 Bcl-xl 的表达情况。结果：在HBE 细胞中转染CIP2A ，A549 细胞中干扰CIP2A ，发现过表达CIP2A 能够显 著抑制肺癌细胞凋亡并上调Bcl-2 和Bcl-xl 的表达，而敲除A549 细胞系中CIP2A 则促进细胞凋亡并抑制Bcl-2 和Bcl-xl 的表达。结论：CIP2A 通过调节Bcl-2 和Bcl-xl 的表达进而影响细胞凋亡。 关键词：病理学；非小细胞肺癌；CIP2A；Bcl-2 ；Bcl-xl ；细胞凋亡 中图分类号：R734.2 文献标识码：A 文章编号：1674-2850(2016)11-1136-06 CIP2A inhibits cell apoptosis in non-small-cell lung cancer DONG Qianze, FU Lin, HAN Yong, XING Jilin (College of Basic Medical Science, China Medical University, Shenyang 110001, China) Abstract: Objective: To investigate whether cancerous inhibitor of protein phosphatase 2A (CIP2A) upregulates the expression of cell apoptosis protein Bcl-2 and Bcl-xl. Cell apoptosis and the expression of Bcl-2 and Bcl-xl were detected using flow cytometry, real-time PCR (RT-PCR) and Western blotting. Results: HBE cell lines were transfected with CIP2A, and A549 cell lines were interfered with CIP2A.
