We use the Electronic Volume technique and measure the Volume directly. Volume increases as the cube of the Diameter, so a small change in Diameter produces a very large change in Volume. A 5% difference is about the limit of the human eye to see and almost impossible to find in 5 cells per hundred We are capable of discerning a less than 2% change in volume which is less than a 1% change in diameter. * Light scatter does a very good job on spheres but on biological cells forward and side scatter measures internal structure as well as diameter and will have a broad distribution not capable of giving the 2% CV you need to determine the NPE. Data obtained from analysis of a 50/50 mixture of 4 micron calibrated spheres and TRBC nuclei using the Electronic Volume channel of the NPE Analyzer (left graph) are compared with Forward Scatter (FSC) data obtained on a Coulter Electronics XL flow cytometer (right graph). The FSC measurement of the spheres does an excellent job. The reverse is true for the biological particles in the mixture.
