多发性硬化患者IFN- 丫及IL220分泌性T细胞的变化 摘要：目的通过计数促炎症性细胞因子IFN- 丫及抗炎症性细胞因子IL-10分 泌性T细胞数目，观察多发性硬化(MS患者此两类细胞因子(CK的变化。 方法采用酶联免疫斑点(Elispot )技术检测MS患者、其他神经疾病(OND患 者及正常对照组(NQ外周血(PB及脑脊液(CSF中MBF及其他抗原反应性 IFN- 丫和IL-10分泌性T细胞数目，并对活动期与缓解期 MSS甲基强的松龙 (MP)冲击疗法前后MS患者PB中两种CK分泌性T细胞数目进行了比较。结果活 动期MS患者PB及CSF中IFN- 丫及IL-10分泌性T细胞数目较OND及 NC组明 显增多(P V 0.05) , CSF中MBP反应性T细胞差异更为显著(P V 0.01)。MS患者 PB中IL-10分泌性T细胞数在缓解期较活动期明显升高(P V 0.05) o MP台疗后 IFN- 丫分泌性T细胞数目明显减少，而IL-10分泌细胞数目明显增多(PV 0.01) o结论在MS发病机制中IFN- 丫起促进作用，而IL-10有保护作用，是 促、抗炎症性CK的比值而非按顺序的表达决定 MS的活动性。MP疗法的部分机 制可能是调节CK网络的平衡。 关键词：多发性硬化； 脑脊液；丫 -干扰素；白细胞介素-10; T细胞 中图分类号： R744.5+1 文献标识码： A 文章编号： 1006-2963 ( 2000 03-146-06 The Changes of IFN- 丫 and IL -10 Secreting T Cells in Multiple Sclerosis Patients ABSTHAC：T ObjectiveTo investigate the changes of cytokines(CK) in multiple sclerosis (MS) by counting the numbers of pro-inflammatory CK IFN- 丫 and anti -inflammatory CK IL-10 secreting T cells. MethodBy reverse Elispot assay,the numbers of the two CK secreting cells stimulated by different antigen in PB and CSF of MS,and other neurological diseases (OND) as well as normal controls (NC) were detected.We also compared the difference of numbers of two subsets of secreting T cell,between the exacerbation and remission stage,before and after methylprednisolone (MP) therapy. ResultsMS patients in the exacerbation stage had more IFN- 丫 and IL -10 secreting T cells in both PB and CSF than OND or NC group(P V0.05),especially when stimulated by MBP and in CSF(P V0.01).In MS remission stage more IL- 10 secreting T cells were found(P V0.05).After MP therapy the number of IFN- 丫 secreting T cells decreased greatly while IL -10 sec-reting T cells in creased(P V 0.01). Con clusio nIFN- 丫 is a disease -promoti ng CK while IL- 10 is a protective CK in MS,it s the ratio between pro - and anti-inflammatory CK that determine the clinical phenomena.The mechanism of MP therapy may be partly due to the balancing effect on the CK network.
