拟南芥Myotubularin2(MTM2)在细胞自噬中的作用研究的中期报告 Abstract: Myotubularin2 (MTM2) is a phosphatidylinositol 3-phosphatase, which plays an important role in regulating autophagy. Previous studies have shown that MTM2 deficiency can lead to autophagy dysfunction and accumulation of autophagic vacuoles in cells. In this study, we aim to investigate the mechanism by which MTM2 regulates autophagy and its potential therapeutic implications. Methods: We used a lentivirus-based shRNA system to knockdown MTM2 expression in HeLa cells. We then assessed autophagy flux using a tandem fluorescent-tagged LC3 reporter assay and western blot analysis of autophagy markers. We also used transmission electron microscopy to examine the morphology of autophagic vacuoles. Results: Our results show that knockdown of MTM2 expression significantly inhibits autophagy flux in HeLa cells, as evidenced by reduced LC3-II levels and accumulation of autophagic vacuoles. We also found that MTM2 knockdown impairs lysosomal function, as indicated by the accumulation of lysosomal substrate and reduced lysosomal acidification. Moreover, we observed that MTM2 knockdown leads to the accumulation of p62, a marker of impaired autophagy flux, and induces ER stress. Conclusion: Our results suggest that MTM2 plays a critical role in regulating autophagy flux and lysosomal function. Our findings also imply that targeting MTM2 may have therapeutic potential for treating autophagy-related diseases. Future studies will explore the potential mechanism by which MTM2 regulates lysosomal function and its potential as a therapeutic target.
