黑果腺肋花楸茎尖培养中不同消毒方法的比较研究 黑果腺肋花楸茎尖培养中不同消毒方法的比较研究 黑果腺肋花楸茎尖培养中不同消毒方法的比较研究 摘 要 本实验以黑果腺肋花楸茎尖为组培材料，研究不同消毒剂和不同处理时间对茎尖消毒效果的影响，使污染降低，提高组织培养的诱导率。本实验分别用浓度为0.1%氯化汞和8%次氯酸钠对黑果腺肋花楸茎尖进行不同时间的消毒处理，然后根据不同消毒剂和不同处理时间对茎尖诱导不定芽进行比较，把诱导率和污染率作为评价指标。结果表明:用8%的次氯酸钠对茎尖进行消毒15min，得苗情况是90%，污染情况为6.67%，褐化情况为3.33%，效果最好;用0.1%氯化汞对茎尖进行消毒6min，得苗情况是3.33%，污染情况为86.67%，褐化情况为10%，效果最差。 关键词:黑果腺肋花楸;茎尖;消毒方法;比较研究 黑果腺肋花楸茎尖培养中不同消毒方法的比较研究 THE ROSACEAE PLANT BLACK CHOKEBERRY SHOOT TIP CULTURE COMPARISON OF DIFFERENT METHODS OF STERILIZATION ABSTRACT The thesis is based on the Rosaceae plant black chokeberry shoot tip as the experimental materials of in use after the different disinfection methods of Sorbus pohuashanensis stem tip regeneration. Sterilization of explants in tissue culture is a very important link, this experiment respectively with 0.1% mercuric chloride and 8% sodium hypochlorite of Sorbus pohuashanensis stem apex for the disinfection of the treatment of different treatment depending on the time of disinfectants and shoot tip induce adventitious buds were compared to induce rate and contamination rate as the evaluation index. Test results show that: with 8% sodium hypochlorite disinfection immersion 15min, The survival rate of shoot tips was 90%, and the pollution was 6.67%, and with the 3.33% browning case, this way was the best results; with 0.1% mercuric chloride were explants disinfection 6min, the survival rate of shoot tips was 3.33%, and the pollution was 86.67%, and browning the case of 10%, this was the worst one.
