Abstract

Theabilitytoregulategeneexpressionisessentialforcontrollingmetaboliceventsinacell.Proteinsthat

functionlikemolecularswitchesrespondtofluctuationsintheenvironmenttomaintainhomeostasis.The

operonmodel,proposedbyJacobandMonod,providesacogentdepictionforhowgeneexpressionis

regulated.Amolecularmechanismfortheregulationfollowedshortlywiththetheoryforallosterictransition.

Overthepasthalf-century,thedetailsofthelacoperonandtheallostericmodelhavebeentestedusing

genetic,biochemical,andstructuraltechniques.Remarkably,theprinciplesoriginallyputforward50years

agoremainessentiallyunchanged.Modelsfortheoperonandthetheoryofallosterictransitionsaretwoofthe

mostprofounddiscoveriesofmolecularbiology.
