现代食品科技 Modern Food Science and Technology 2008, Vol.24, No.11 泰山灰树花液体深层发酵培养基的优化及 海藻糖的提取 李艳玲,苗苗,王健美,史仁玖,李恒 (泰山医学院生物科学系,山东 泰安 271016) 摘要:本文对泰山灰树花液体发酵培养基的配方进行了优化,并对灰树花中海藻糖的测定条件和提取工艺进行了初步研究。结 果表明,优化的发酵培养基组成为:马铃薯200 g/葡萄糖40 g ,麸皮25 g ,板栗壳 150 g,磷酸二氢钾3 g,硫酸镁 1.5 g,水 1 L,菌 n 丝体生物量为 15.1 g/L。用0.2%蒽酮-硫酸溶液(料液比 1:4 )测定菌体中海藻糖含量的条件:吸收波长625 nm,加热时间2 min。海 c 藻糖提取的工艺参数是:冰水浴,0.5 mol/L 三氯乙酸 20 mL ,提取次数为 3 次,每次提取时间 20 min ,共从 1.0 g 湿菌丝中提取到 14.522 . mg 海藻糖。 关键词:灰树花;发酵培养基;蒽酮-硫酸法;海藻糖 m 中图分类号:TS201.3;文献标识码:A ;文章篇号:1673-9078(2008)11-1141-04 o c Optimization of Fermentation Culture Medium for Grifola frondosa d. and Extraction of Trehalose o LI Yan-ling, MIAO Miao,WANG Jian-mei, SHI Ren-jiu, LI Hen (Department of Biological Sciences, Taishan Medical College, Taian 271016, China) o f Abstract: The fermentation medium composition of Grifola frondosa was optimized, and the extraction of trehalose from Grifola frondosa was studied.
