D ep a rtment of S u rg ery Abstract :Objective 　To exp lore t he effect s on chemot herap eutic sen sitivit y of gast ric cancer cell line by st able overexp ression of smac gene . Methods 　U nder t he induction of lipo some , t he eukaryotic exp res sion vector p cDN A3 . 1smac for smac gene and it s cont rol vector p cDN A3 . 1 were t ran sfect ed into ga st ric cancer cell line M KN4 5 . The subclone cell lines were obt ained by p er si st ent G selection . smac gene 4 18 exp ression of cancer cell s were det ect ed by R TPCR and West ern Blot met ho ds. The growt h inhibition effect s of mitomycin (MMC) on cancer cell s were al so ob served by t et razolium bromide colorimet ry and clone formation t est . Results 　The subclone ga st ric cancer cell lines , st able exp ressing smac and neo gene resp ectively , were successfully select ed , named as M KN4 5/ smac and M KN45/ neo .
