Abstract:Objective:StudyonmechanismofactionoftraditionalChinesepreparationZhixiaotongmaininginrenal
interstitialfibrosis(RIF)ofmousewithdiabeticnephropathy.Methods:Fiftyhealthy12-week-oldmaleKKAymiceandten
C57BL/612-week-oldmicewererandomlydividedintoVehiclegroup,modelgroup,ValsartangroupandZhixiaotongmalning
(ZXTMNN)high-,medium-,low-dosegroup.Thenwerecordedthegeneralstateofthemice,detectedthebloodglucose,
urineproteinquantification,Scrin24h,levelsofserumcreatinine(Scr)andMassonstainingofkidney;wedetectedtheprotein
expressionsofα-SMA,TGF-β1,BMP-7andVEGFofnephridialtissuewithWesternBlot;andanalyzedtheexpressionofTGF-β1
mRNA,TGF-βR1mRNAandSmad2mRNAofnephridialtissuewithReal-timeRT-PCR.Results:ZXTMNandvalsartancould
dropbodyweight,bloodglucoseandthe24hurineprotein.Thequantitativeandsemi-quantitativeanalysisofRIFresultsofthe
miceweresignificant(P＜0.05)inthefourthweek.Theproteinexpressionsofα-SMAandTGF-β1ofthemiceintheValsartan
group,high-andmedium-doseZXTMNgroupsdroppedsignificantly(P＜0.01).AndtheproteinexpressionsofBMP-7andVEGF
bothincreasedsignificantly(P＜0.01).TheexpressionsofTGF-β1mRNAandSmad2mRNAinthehigh-,medium-and
low-ZXTMNgroupsdroppedsignificantly(P＜0.01).Conclusion:RegulatingrenalTGF-β1/Smadsignalpathwaymightbeoneof
themechanismsofZXTMNtherapeuticeffectofDN.
