降钙素对兔骨关节炎关节软骨的保护作用_医学论文 降钙素对兔骨关节炎关节软骨的保护作用_医学论文 【关键词】 降钙素 　　摘 要：［目的］研究降钙素(calcitonin，CT)对骨关节炎关节软骨的保护作用。［方法］6个月龄新西兰大白兔20只，随机分为两组均行右膝关节前交叉韧带切断术。1周后实验组开始皮下注射降钙素5 IU／kg，每日1次；对照组注入等剂量生理盐水；术后6周处死动物。取股骨内髁制成切片行HE、番红“O”和Ⅱ型胶原免疫组化染色。HE切片按照Mankin评分表评分。番红“O”染色和Ⅱ型胶原免疫组化染色切片用图像分析仪，测量平均灰度值。［结果］(1)对照组术侧膝关节组织学Mankin评分结果、番红“O”染色和Ⅱ型胶原免疫组化染色平均灰度值测量结果高于健侧，（001）。（2)对照组手术膝关节组织学评分、番红“O”染色和Ⅱ型胶原免疫组化染色平均灰度值测量结果高于实验组手术膝关节，（005）。［结论］降钙素5 IU/ kg每日1次皮下注射够明显减轻兔膝关节不稳定诱发的骨关节炎关节软骨的退变。降钙素可能通过提高软骨基质糖胺多糖(glycosaminoglycans，GAG)和Ⅱ型胶原含量保护关节软骨。 　　关键词：降钙素； 骨关节炎； 灰度值 　　Abstract：［Objective］To assess whether calcitonin exerts an chondroprotective action in unstable knees which induce experimental osteoarthritis. ［Method］Twenty 6month New Zealand rabbits were divided into two groups randomly. All rabbits underwent anterior cruciate ligament transection (ACLT) on the right knee. Rabbits of experimental group began to receive a daffy subcutaneous injection of salmon calcitonin at a dose of 5 IU per kg of body weight on day 7 postsurgery in contrast ,control group received normal saline (NS) at the same dose. All rabbits were killed on day 42 postsurgery. The knee joints were examined grossly and histologically and scored the medial condyle sections were stained with Safranin O and immunohistochemistry of collagen II. We applied a computer graphic technology to assess the glycosaminoglycans (GAG) and collagen II of articular cartilage. ［Result］(1) In control group, compared uninjured knee joints with operative knee joints,the appearance of operative knee joints accorded with OA and histologic score of operative knee joints were higher than that of uninjured knee joints,0.01 the grey value of Safranin O and collagen II of operative knee joints were greater, too.(2) Compared control group with experimental group,the histologic score, the grey value of Safranin O and collagen Ⅱ control group were higher than that of experimental group. ［Conclusion］ A daily subcutaneous injection of salmon calcition at a dose of 5 IU/k
