[Abstract] Objective：Duchennemusculardystrophy(DMD)andBeckermusculardystrophy(BMD)are
X—linkeddisorderscausedbymutationsinthedystrophingene．Themajorityofrecognizedmutationsare
copynumberchangesofindividualexons．Theobjectiveofthepresentstudywastoassessthemultiplex
ligation—dependentprobeamplification (MLPA)effectsofdetectionofgenemutations．M ethods：Samples
of20controlmalesand80malesandtheirmothersreferredtoourdiagnosticfacilityontheclinicalsuspi—
cionofDMDorBMDweretestedbyMLPAandmultiplexPCR．Results：ThemeanDQsforallpeakof20
controlmalesampleswas1．02(rangefrom 0．83to 1．21)byM LPA．Deletionsorduplicationswereiden—
tifiedin 6outof31familiesthathadbeenpreviouslytestedasnegativebymultiplex PCR．Onecaseof
complexrearrangementinvolvingaduplicationoftworegions：dupEX3—9anddupEX 17-41werefoundby
MLPA．Conclusions：MLPA isahighlysensitivemethodandrapidalternativetomultiplexPCR fordetec—
tionofDMD andBMD．
