Cyclin D_1在颅脑胶质瘤中表达及细胞周期表达模式的研究 　　叶元胜 　詹仁雅 　童 　鹰 　　作者单位:325604 浙江省乐清市第三人民医院(叶元胜) 　　310003 浙江大学医学院附属第一医院(詹仁雅 童鹰) 　　【摘要】　 目的 研究颅脑胶质瘤中cyclin D1的表达和细胞周期表达模式,探讨其与胶质瘤恶性程度的关系,并从生物学功能上探讨Cy2clinD1在胶质瘤细胞周期演进中的作用。 方法 采用免疫组化法检测52例胶质瘤标本中cyclin D1 的表达,观察其与胶质瘤恶性程度之间的关系;利用流式细胞仪双参数法和细胞同步化,确定 CyclinD1在胶质瘤细胞系SHG- 44和BT - 325中的细胞周期表达分布模式。 结果 　Cyclin D1的表达随病理等级的递增而增高,高恶性度组阳性率和标记指数明显高于低恶性度组( P 0. 05) 。在胶质瘤细胞系 SHG- 44 和BT - 325中,CyclinD1的表达呈细胞周期依赖性。 结论 　 cyclin D1在人脑胶质瘤细胞中过度表达,同肿瘤的发生、发展有密切联系;在SHG-44和BT - 325胶质瘤细胞中 CyclinD1的表达呈细胞周期依赖性。 　　【关键词】　 细胞周期素D　免疫组化　胶质瘤　细胞周期 　　【Abstract】　Objective 　To investigate the expression and expression model of cyclin D1 in human glioma and reveal their relationships with pathological malignancy grades and elucidate biological function of cyclin D1 in the progression of cell cycle. Methods　Immunohistochemistry was used to detect the expression of cyclin D1 in 52 glioma cases in order to reveal their relationships with pathological malignancy grades. Cyclin D1 expression model in glioma cell line SHG- 44 and BT - 325 was analysed by using double parameter flow cytometry and cell synchronization. Results　 Marked overexpression of cyclin D1 was observed in gliomas. Expression of cyclin D1 increased with advancing malignancy grades. Positive rates and labeling indices in higher malignancy grades were significantly higher than those in lower malignancy grades ( P 0. 05) . Double parameter flow cytometry of SHG- 44 and BT - 325 showed that Cyclin D1 expression was manifested as cell cycle dependent manner and oscillated with cell cycle. Conclusion　 Cyclin D1 are over - expressed in human gliomas and closely related to oncogenesis and progression of gliomas.
