三种不同染色方法对黄瓜白粉病菌的染色观察 任红敏 杨军玉 王树桐 曹克强 河北农业大学植物保护学院，河北省农作物病害生物防治工程技术中心，保定071001 摘要:比较了考马斯亮蓝、曲利苯蓝、苯胺蓝应用于黄瓜白粉病菌染色的效果。结果表明,考马斯亮蓝组织透明染 色方法和曲利苯蓝组织透明染色方法都可以清楚观察到白粉病菌的5个发育阶段，即萌发的分生孢子初生芽管、附 着胞、胞间菌丝、分生孢子梗和后期菌落。考马斯亮蓝组织学染色方法样品处理过程需要35min左右，染色后使寄 主组织几乎不着色，病菌菌体染成深蓝色，染色迅速，仅需1～2min;曲利苯蓝染色法样品处理过程仅需要25min 左右，寄主组织脱色彻底，呈浅蓝色，菌体染色后为深蓝色，与背景反差大，便于观察，且使用药品的配制相对简 便；苯胺蓝组织透明染色方法也可观察到病菌的不同发育阶段，但观察效果不理想，并且整个处理过程需要耗时24h 之多。 关键词:黄瓜白粉病菌；组织透明观察 ComparisonofStainingEffectsofThreeStainingTechniquesfor ObservationofSphaerothecafuliginea RENHong-minYANGJun-yuWANGShu-tongCAOKe-qiang CollegeofPlantProtection,Bio-controlCenterofPlantDiseaseandPestsofHebeiProvince,AgriculturalUniversityof Hebei,Baoding071001,China Abstract:StainingeffectsofCoomassiebrilliantblue,TrypanblueandAnilineblueonSphaerothecafuligineaPollwere observed.TheresultsshowedthatbothCommassiebrilliantbluestainingandTrypanbluestainingcanclearlydisplaythe differentstructuresofSphaerothecafuliginea,suchasgerminatingconidia,initialgermtubes,appressorium,hyphae, conidiophoresandlargecolony.Commassiebrilliantbluestainingneedabout35minofsampletreatment,andstain pathogendarkbluerapidlywhichonlyneed1～2min,whilehardlystainthehosttissue.Trypanbluestainingneedonly25 minofsampletreatment,andthehostcellturnedintolightblueafterdecolorizedcompletely,andpathogendarkblue,this gaverisetostrongcontrastforobservation,meanwhilethepreparationofdrugsisrelativelysimpleforthismethod.Aniline bluestainingcanalsodisplaythedifferentstructuresofSphaerothecafuliginea,buttheobservationresultisnotsatisfactory, moreover,sampletreatmentneedmorethan24h.
