ChinMedSciJ V01．26．No．4 December2011 P 216—22O CHINESE MEDICALSCIENCES JOURNAL oRIGINAL A TICLE ContraryRegulationofTIMP一1andMMP一9byHepatocyte GrowthFactorAntibodyafterLungInjury Wei-weiYu，andQinXia nstituteofGeneralDepartment，TongjiHospital，TongjiMedicalCollege，Huazhong UniversityofScienceandTechnology，Wuhan430030，China Keywords：hepatocytegrowthfactor；matrixmetalloProteinases一9；tissueinhibitorof metalloProteinases一1 Objective Tostudytheinfluenceofhepatocytegrowthfactor(HGF)antibodyonthelungexpres— sionlevelofmatrixmeta110proteinases一9(MMP一9)andtissueinhibitorofmetalloPr0teinase一1(TIMP一1)． Methods ThirtymaleW istarratswererandomlydividedinto3groups：controlgroup，modelrgoup， andinterventionrgoup．Endotoxinwasintratracheallyinfusedinthemodelandinterventiongroups．HGF antibodywasinjectedintheratsofhteinterventiongroupfrom day1today14，whilethesamevolumeof salinewasinjectedinthecontrolrgoup．Theratsweresacrificedonday28afterendotoxintreatment．The amountsofMMP—-9mRNAandTIMP—_1mRNAweremeasuredbyreversetranscription—polymerasechain reaction，andproteinexpressionlevelsofMMP一9andTIMP一1weremeasuredbyimmunohistochemistry． Results Inhtemodelgroup，bohtmRNAandproteinexpressionlevelsofTIMP一1weresignificantly increased，htesameasMMP一9．Intheinterventiongroup，hteincreaseofTIMP一1wasremarkablyreduced comparedwithhtemodelgroup，whilehtemRNA andproteinexpressionlevelsofMMP一9werestillin— creased．
