促红细胞生成素兔视网膜神经节细胞的保护作用.pdf

摘 要 目的：建立兔眼慢性高眼压模型，探讨促红细胞生成素（EPO）对兔视网膜神经节 细胞的保护作用。 方法：新西兰大白兔35 只，随机分3 组，I 组即正常对照组5 只，II 组即促红素 玻璃体注射组15 只和III 组即损伤组15 只，II 和III 组左眼前房注射卡波姆眼液制成 慢性高眼压模型，schiotz 眼压计测量眼压。II 组制模成功后开始向玻璃体腔注射 rHuEPO 3uL 3000U/ml，1 次/wk，连续3 周。每组取10 只眼球固定后行石蜡切片，做 HE 染色及TUNEL 染色检测各组实验结果。 结果：400 倍光镜下，HE 染色观察视网膜神经节细胞计数，I 组正常组视网膜神经 节细胞计数是37.21±0.83；Ⅱ组治疗组模型眼视网膜神经节细胞，计数是29.82±0.84； Ⅲ组损伤组模型眼视网膜神经节细胞减少，计数是25.35±0.76；与正常对照I 组相比 较，II 和III 组模型眼的视网膜神经节细胞数均明显减少，差异具有统计学意义 （P0.05）；III 组即损伤组模型眼与II 组治疗组比较，视网膜神经节细胞数减少，具 有显著差异（P0.05）；I 组TUNEL 染色阳性细胞数0.27±0.19；II 组TUNEL 染色阳性 细胞数3.99±0.26；III 组TUNEL 染色阳性细胞数10.82±0.98；TUNEL 染色II 和III 组模型眼视网膜神经节细胞凋亡数均高于对照眼，差异具有统计学意义（P0.05）；II 组模型眼TUNEL 染色视网膜神经节细胞凋亡数少于III 组模型眼，差异具有统计学意义 （P0.05）。 结论：促红细胞生成素对兔慢性高眼压模型的视网膜神经节细胞有保护作用。 关键词：青光眼，视网膜神经节细胞，EPO 5 Abstract Objectiv To observe the neuroprotection of erythropoietin on rabbit retinal ganglion cells in chronic ocular hypertention model ． Methods 35 rabbits without eye diseases were divided into 3 groups ：groupI ：normal group(5rabbits) ，groupII ：erythropoietin treated group(15 rabbits) ，groupIII ：balanced salt solution treated group(15 rabbits) ．Rabbits in group II and III were induce III left eye by forcing pefusion of balanced salt solution into the anterior chamber as chronic ocular hypertenfion model ．Right eyes were self-control eyes ．Not standard eyeballs were discard andl0 eyeballs were selected and fixed in each group ．(The retinal ganglion cells were detected by HE staining .The apoptosis cells were detected by TdT-dUDP terminal nick end-labeling (TUNEL)technique ． Results The RGCs quantity of model eyes in group II and III were less than that of group I and self-control eyes ，there were significant diferences(P0.05) ．But the RGCs quantity of model eyes in groupII were more than that of groupIII ，there were significant difere
