HPLC梯度洗脱法同时测定川芎中阿魏酸和藁本内酯的含量 汪潜1，朱玉丰1，李鹏2 ，王一涛2 （1．太极集团有限公司，重庆 401147； 2. 澳门大学，澳门） 【摘 要】 目的：建立HPLC梯度洗脱的方法同时测定川芎中阿魏酸和藁本内酯的含量，以评价川芎药材的质量。方法：采用ODS C18 （4.6×250 mm，5μm）色谱柱，乙腈-水（1% 醋酸）为流动相进行梯度洗脱，流速为1.0ml. min-1 ，检测波长为284nm。结果：在该条件下，阿魏酸和藁本内酯分离良好，阿魏酸在0.102～1.632μg范围内线性良好（r 0.9999），平均回收率为100.2%，RSD为1.0%；藁本内酯在0.4～4.0μg范围内线性良好（r 0.9999），平均回收率为98.4%，RSD为1.0%。结论：该方法分离效果好，简便快捷。 【关键词】 川芎； 阿魏酸； 藁本内酯； 梯度洗脱； HPLC Determination of Ferulic Acid and Z-Ligustilide in Ligusticum Chuanxiong Hort by HPLC Wang Qian1, Li Peng1 , Kelvin Kan1，Li Shao-ping1, Wang Yi-tao1,2 （1．University of Macau，Macau；2．China Academy of Traditional Chinese Medicine，Beijing 100700 【Abstract】 Objective: To establish a gradient elution method for simultaneous determination of ferulic acid and ligustilide in Ligusticum chuanxiong Hort , so as to evaluate the quality of the Rhizome Chuanxiong . Method: ODS C18 column（4.6×250 mm，5μm）was used. The mobile phase for gradient elution consist of acetonitrile and water 1%HAc , the flow rate was 1.0 ml. min-1, the wavelength for measurement was 284nm. Results: In this condition, ferulic acid and ligustilide can be successfully separated, the ferulic acid has a good linear relation at the range of 0.102～1.632μg（r 0.9999）, the average recovery was 100.2%, RSD was 1.0%, the ligustilide has a good linear relation at the range of 0.4～4.0μg （r 0.9999）, the average recovery was 98.4%, RSD was 1.0% . Conclusion: This HPLC method is a simple, rapid method for simultaneous determination of ferulic acid and ligustilide in the Ligusticum chuanxiong Hort. 【Key words】 Ligusticum chuanxiong Hort; ferulic acid; ligustilide; gradient elution; HPLC 川芎为伞形科藁木属植物Ligusticum chuanxiong Hort.干燥的根茎。性温，味辛，具有活血行气，祛风止痛的功能[1]；用于月经不调、经闭痛经、产后瘀滞腹痛、胸肋刺痛、跌扑肿痛、头痛、风湿痹痛[2]，是一种常见的中草药。川芎含有多种化学成分[3-4]，有效成分可以分为水溶性和脂溶性两部分，分别以阿魏酸[5]和藁本内酯[6]为代表。因此，研究与检测制剂中阿魏酸及藁本内酯的含量，对于保证疗效及提高产品质量有积极意义。 阿魏酸和藁本内酯两种成分的极性相差较大，目前尚无同时测定两种成分的方法，于是在本实验中，我们通过参考文献[7]，考虑采用梯度洗脱的方式，将乙腈和水按不同的比例，以及调节水相的PH值来选择梯度洗脱的条件，最终确定了乙腈-
