* * To perform a one-stage assay, citrate, which acts as an anticoagulant by binding calcium, is added to blood to inhibit coagulation until assay begin. The citrated blood is mixed and centrifuged to isolate the plasma component. Thromboplastin, a recombinant or crude animal tissue extract that provides phospholipids and TF, is then added, followed by calcium, which neutralises the citrate and facilitates clot formation, and the time to fibrin clot formation is optically assessed.
