温度对黑线仓鼠抗氧化能力和解偶联蛋白表达的影响

摘要:根据自由基的假说,高代谢率应该会增加活性氧(ROS)的产生。然而,“解偶联生存”假说认为解偶联蛋白(UCPs)ROS的产生,在这种情况下,代谢速率和ROS的产生之间的关系将是负相关而不是正相关。在本研究中,我们研究黑线仓鼠能量摄入、氧化应激水平、抗氧化活性,以及褐色脂肪组织(BAT)、肝脏、心脏、骨骼肌和大脑解偶联蛋白的表达。将黑线仓鼠驯化于5℃和32.5℃的环境,而对照组饲养在室温下 (21°C)。相对于对照组,在5°C环境中驯化的黑线仓鼠能量摄入增加了70.7%,而在32.5°C环境中驯化的黑线仓鼠能量摄入减少31.3%。相对于对照组,在5°C环境中驯化的黑线仓鼠的丙二醛(MDA)水平、总抗氧化能力(T-AOC)和谷胱甘肽过氧化物酶(GSH-PX)在BAT中显著降低。相对于对照组,在5°C环境中驯化的黑线仓鼠的UCP1在BAT中的表达显著上升,而在32.5°C环境中显著下降。在5°C环境中驯化的黑线仓鼠其UCP3在骨骼肌的表达也大幅上升。这些结果表明,活性氧的产生和代谢率之间的关系是负相关,而不是正相关。

Barabensis temperature oxidation resistance and uncoupling protein expression

Abstract: In line with living-free radical surmise, higher metabolic rates should subjoin reactive oxygen species (ROS) production. But, the “uncoupling survival” surmise assume that uncoupling proteins (UCPs) by reducing mitochondrial membrane potential to lessen the operation of ROS, in this case, the relationship between metabolic rate and ROS production will be a negative correlation between rather than positive. We barabensis into 5 °C and 32.5 °C environment domesticated. The control group was 21 °C. Compared with the control group, the energy intake at 5 °C domesticated barabensis increased by 70.7%, while the black line at 32.5 °C hamster domestication decreased by 31.3%. Malonadialdehyde (MDA) levels, total antioxidant capacity (T-AOC) and glutathione peroxidase (GSH-PX) activity in BAT significantly decreased in hamsters acclimated to 5°C, but increased in those acclimated to 32.5°C, relative to the control. Neither ROS production, nor antioxidants in skeletal muscle, liver, heart or brain tissue, were affected by temperature. UCP1 expression in BAT was significantly up-regulated in hamsters acclimated to 5°C, but down-regulated in those acclimated to 32.5°C, relative to the control.
