摘 要 马铃薯是一种重要的粮菜兼用作物,同时它营养成分全面,营养结构合理,易于消化吸收,是世界十大营养食品之一.在马铃薯的块茎中含有淀粉、蛋白质、脂肪、粗纤维等营养成分以及胡萝卜素、抗坏血酸、酚酸等抗氧化物质.本文研究了马铃薯中酚酸物质—绿原酸的提取和纯化工艺.得到了相应的工艺参数. 采用超声波提取法对马铃薯中的绿原酸进行提取.适宜的提取工艺参数:pH = 2,70%的乙醇溶液;料液比为1:8;提取温度:40℃;提取时间:30 min;提取次数:2 次;提取功率:160 W. 利用紫外-可见吸收光谱法(Ultraviolet and Visible Spectrometry,UV-VIS)对马铃薯中的绿原酸进行定性、定量检测.通过扫描标准品的紫外光谱曲线,确定绿原酸的最大吸收波长是330 nm;在不同的提取条件下,测定绿原酸提取液的吸光度可以确定最佳提取条件. 利用高效液相色谱法(High Performance Liquid Chromatography,HPLC)进行绿原酸的定性分析.通过观察粗提液以及粗提液与标准品混合溶液的保留时间,得到的实验结果为:粗提液中确实存在绿原酸,且两种溶液的保留时间几近相同,绿原酸峰两侧的杂峰很小,可忽略不计. 采用树脂吸附法进行纯化.分别采用3种树脂:NKA-9型树脂(强极性)、AB-8型树脂(弱极性)以及 D101型大孔树脂(非极性).通过静态吸附和解吸附确定了NKA-9型树脂具有较好的吸附和解析附效果,饱和吸附量为:6.85 mg/g,解吸率为:70.8%.;通过动态吸附和解吸附描绘出的动态饱和吸附和解吸附曲线,可以知道,NKA-9型树脂能够达到较好的饱和吸附与解吸附效果. 关键词: ABSTRACT The potato is a kind of important food dish saut and industrial crops, at the same time, it has the comprehensive nutrients as well as the reasonable structure of nutrition, the potato is one of the world’s top ten nutrition food which is easy to digest and absorb. There are starch, protein, fat, crude fiber nutrients in the potato tuber. Also, carotene, ascorbic acid, phenolic acids and other antioxidants were included. This paper studies the matter- the extraction and purification process of chlorogenic acid from potato phenolic acids. Through the experiment, I have got the corresponding technological parameters. We use the ultrasonic wave assisted extraction to extract the chlorogenic acids. The best technological parameters as follows: pH = 2, 70% ethanol as extraction solvent; tatio of material to liquor 1:8, the ultrasonic wave power 160W, the extraction temperature 40℃, the time for 30 min, to extract 2 times. Using the Ultroviolet and Visible Spectrometry (UV-VIS) to determine the qualitative and quantitative detection on the chlorogenic acid of the potato, we conform the maximum adsorption wavelength of chlorogenic acid is 330 nm through the Ultroviolet spectrum curve.In different extraction conditions, we can determine the best extract
