The Origin of the Neoplastic Stromal Cell in Giant Cell Tumor of Bone MANUELA WU¨ LLING, PHD, GUENTER DELLING, MD, AND EDELGARD KAISER, PHD Fibroblastlike stromal cells, which are always present as a component of giant cell tumor of bone (GCT), can be observed in both in vivo and cultured cell samples. Although they are assumed to trigger the cancer process in GCT, the histogenesis of GCT stromal cells is poorly understood. It is known that mesenchymal stem cells (MSCs) can develop to osteoblasts. Evidence has been presented that GCT stromal cells can also develop to osteoblasts. A connection between MSCs and GCT stromal cells was sought by using 2 different laboratory approaches. First, immunohistological analyses revealed that some of the same markers, detected by the SH2, SH3, and SH4 antibodies and the CD166 antigen, were found in GCT stromal cells as in the ?rst developmental stages of osteoblast differentiation from Mesenchymal stem cells (MSCs) are multipotent cells that reside in the bone marrow. They are capable of differentiation into osteoblasts, chondroblasts, myoblasts, myelosupportive stromal cells, and adipocytes in the respectively appropriate environment in which growth signals are rendered. Although their histogenesis has not been fully elucidated,1 in animal studies, MSCs were able to regenerate bone and bone marrow on in vivo transplantation.2,3 Stem cell research has bounded ahead in recent years, allowing new conclusions about cell development. A methodology for the isolation and cultivation of human MSCs has been in development since the 1960s.4 Since 1991, a selection of antibodies (SH2, SH3, SH4, SB10/ALCAM/CD166, Stro1) has been available with which the observation of the differentiation process of MSCs into osteoprogenitors and preosteoblasts is possible, thereby completing the developmental chain of the osteogenic lineage into osteoblasts.5-8 Knowing the markers of an individual cell allows staging of that cell in terms of its sequential
