Plant preparation of superoxide dismutase Technology

[Abstract] Objective To optimize the plant superoxide dismutase (SOD) of the preparation process. Methods thermal denaturation precipitation, isoelectric point precipitation, 45%, 90% ammonium sulfate fractionation, DEAE-cellulose ion-exchange chromatography, freeze drying methods such as concentration, from the Preparation of cucumber SOD, and used polyacrylamide gel electrophoresis identification. With Coomassie Brilliant Blue G-250 Determination of protein content, using NBT photoreduction Determination of SOD activity. Results SOD enzyme specific activity as 89.2381 U * mg-1, recovery was 18.41%, purification multiples of 11.02 times. Conclusion The process is simple and efficient purification from the plant superoxide dismutase, can be used for actual production.

Abstract: ObjectiveTo optimize the extraction technology of superoxide dismutase from plants. MethodsWe extracted SOD from the cucumber by heat treatment, isoelectric fraction, ammonium sulfate fractionation and ion exchange chromatography, Coomassie brilliant blue G-250 was adopted to determine the content of the protein and NBT-reduction was used to measure the SOD activity. Meanwhile, the purified substance was identified as SOD by the polyacrylamide gel electrophoresis (PAGE) and SOD activity staining. ResultsSOD had a specific activity of 89.2381 U * mg-1. The yield rate was 18.41%, and the multiplication was as 11.02 times. ConclusionThe technology is an extraction technology of superoxide dismutase form plants simply and efficiently and it will be a good reference for the production.
