Despiteadvancesinsurgery,radiationtherapy,andchemotherapeutics,patientswithmalignantgliomahaveadismalprognosis. Theformationsofaberranttumourvasculatureandgliomacellinvasionaremajorobstaclesfore?ectivetreatment.Angiogenesis isakeyeventintheprogressionofmalignantgliomas,aprocessinvolvingendothelialcellproliferation,migration,reorganization ofextracellularmatrixandtubeformation.Suchprocessesareregulatedbythehomeostaticbalancebetweenproangiogenicand antiangiogenicfactors,mostnotablyvascularendothelialgrowthfactors(VEGFs)producedbygliomacells.Currentstrategies targeting VEGF-VEGF receptor signal transduction pathways, though e?ective in normalizing abnormal tumor vasculature, eventuallyresultintumorresistancewherebyahighlyin?ltrativeandinvasivephenotypemaybeadopted.Herewereviewrecent anti-angiogenic therapy for malignant glioma and highlight implantable devices and nano/microparticles as next-generation methods for chemotherapeutic delivery.
