Malignantgliomashaveapoorprognosisdespiteadvancesindiagnosisandtherapy.Althoughpostoperativetemozolomideand radiotherapyimproveoverallsurvivalinglioblastomapatients,mostpatientsexperiencearecurrence.Theprognosisofrecurrent malignantgliomasisdismal,andmoree?ectivetherapeuticstrategiesareclearlyneeded.Antiangiogenesisiscurrentlyconsidered anattractivetargetingtherapyformalignantgliomasduetoitsimportantroleintumorgrowth.Clinicaltrialsusingbevacizumab havebeenperformedforrecurrentglioblastoma,andthesestudieshaveshownpromisingresponseratesalongwithprogression- freesurvival.Basedontheencouragingresults,bevacizumabwasapprovedbytheFDAforthetreatmentofrecurrentglioblastoma. Inaddition,bevacizumabhasshowntobee?ectiveforrecurrentanaplasticgliomas.LargephaseIIIstudiesarecurrentlyongoing todemonstratethee?cacyandsafetyoftheadditionofbevacizumabtotemozolomideandradiotherapyfornewlydiagnosed glioblastoma. In contrast, several other antiangiogenic drugs have also been used in clinical trials.
