ThemicrobiotaofthegastrointestinaltractisfrequentlymentionedasoneofthekeyplayersintheetiopathogenesisofCrohn’s disease(CD).Fourhypotheseshavebeensuggested:thesingle,stillunknownbacterialpathogen,anabnormaloverallcomposition ofthebowelmicrobiota(“dysbiosis”),anabnormalimmunologicalreactiontoanessentiallynormallycomposedmicrobiota, andincreasedbacterialtranslocation.Weproposethatlasercapturemicrodissectionofselectedmicroscopicstructures,followed bybroad-range16SrRNAgenesequencing,isanexcellentmethodtoassessspatiotemporalalterationsinthecompositionof the bowel microbiota in CD. Using this approach, we demonstrated signi?cant changes of the composition, abundance, and locationofthegutmicrobiomeinthisdisease.Someoftheseabnormal?ndingspersistedevenaftermacroscopicmucosalhealing.
