Atopicdermatitisisamultifactorial,chronicrelapsing,in?ammatorydisease,characterizedbyxerosis,eczematouslesions,and pruritus.Thelatterusuallyleadstoan“itch-scratch”cyclethatmaycompromisetheepidermalbarrier.Skinbarrierabnormalities inatopicdermatitismayresultfrommutationsinthegeneencodingfor?laggrin,whichplaysanimportantroleintheformation ofcorni?edcytosol.Barrierabnormalitiesrendertheskinmorepermeabletoirritants,allergens,andmicroorganisms.Treatment ofatopicdermatitismustbedirectedtocontroltheitching,suppressthein?ammation,andrestoretheskinbarrier.Emollients, bothcreamsandointments,improvethebarrierfunctionofstratumcorneumbyprovidingitwithwaterandlipids.Studieson atopicdermatitisandbarrierrepairtreatmentshowthatadequatelipidreplacementtherapyreducesthein?ammationandrestores epidermalfunction.E?ortsdirectedtodevelopimmunomodulatorsthatinterferewithcytokine-inducedskinbarrierdysfunction, provideapromisingstrategyfortreatmentofatopicdermatitis.Moreover,animpressiveproliferationofmor
