Brainmetastasis,animportantcauseofcancermorbidityandmortality,occursinatleast30%ofpatientswithbreastcancer.A keyeventofbrainmetastasisisthemigrationofcancercellsthroughtheblood-brainbarrier(BBB).Althoughpreventingbrain metastasisisimmenselyimportantforsurvival,verylittleisknownabouttheearlystageoftransmigrationandthemolecular mechanisms of breast tumor cells penetrating the BBB. The brain endothelium plays an important role in brain metastasis, althoughthemechanismsare notclear. BrainMicrovascularEndothelialCells (BMECs)are themajorcellularconstituentof theBBB.BMECsarejoinedtogetherbyintercellulartightjunctions(TJs)thatareresponsibleforacquisitionofhighlyselective permeability.FailureoftheBBBisacriticaleventinthedevelopmentandprogressionofseveraldiseasesthata?ecttheCNS, includingbraintumormetastasisdevelopment.Here,wehavedelineatedthemechanismsofBBBimpairmentandbreastcancer metastasistothebrain.UnderstandingthemolecularmediatorsthatcausechangesintheBBBshouldleadtobetterstrategiesfor e?ectivetreatmentmodalitiestargetedtoinhibitionofbraintumors. 1.Introduction andad
