Methodology/PrincipalFindings:Usingconventionalantibodiesandinmunocytochemistry,differentialcentrifugationand westernblot,wehaveobservedtheputativepresenceofK-Rasisoforminthenucleioffibroblastsandmesangialcells.In ordertoavoidcross-reactionswithotherRasisoforms,andusingantibodiesagainstK-Ras(R-3400,H3845-M01,sc-30)or pan-Ras(05-516,OP40)incellsthatonlyexpressedtheK-Rasisoform(fibroblastsobtainedfromH-ras2/2,N-ras2/2mice)we alsodetectedsomenuclearpositiveexpression.TofurtherprobetheidentityofnuclearK-Ras,wehavegeneratedK-Ras knockout(K-ras2/2)embrionaryfibroblastsbymatingofK-ras+/2heterozygotemice.Usingspecificantibodies,onlyH-and N-RasisoformswereobservedinthecytoplasmofK-ras2/2fibroblasts.However,bothK-Ras4AandK-Ras4Bpositivesignals weredetectedbyimmunocytochemistryandWesternblotwithtwocommercialantibodies(sc-522andsc-521againsteach isoforms,respectively)inbothcytoplasmandnucleifromK-ras2/2 fibroblasts. Conclusions/Significance:WeshowthatthepresenceofK-Ras4Binfibroblastnuclei,alreadydescribedbyotherauthors,is probablyduetoacross-reactionoftheantibodywithanundeterminednucleolarprotein.Althoughthisstudyalsoshows thepossiblenuclearexpressionofK-Rasisoforminfibroblastsorinmesangialcells,italsorevealstheimportanceofbeing cautious in these stu
