Asthec-Myconcoproteinhasrecognizedrolesinnormalstemcellbiology,wehypothesizedthatc-Mycmaycontributeto cancerstemcellbiologyasthesecellssharecharacteristicswithnormalstemcells. Methodology/PrincipalFindings:Basedonpreviousmethodsthatweandothershaveemployed,tumorcellpopulations wereenrichedordepletedforcancerstemcellsusingthestemcellmarkerCD133(Prominin-1).Wecharacterized c-Myc expression in matched tumor cell populations using real time PCR, immunoblotting, immunofluorescence and flow cytometry.Herewereportthatc-Mycishighlyexpressedingliomacancerstemcellsrelativetonon-stemgliomacells.To interrogatethesignificanceofc-Mycexpressioningliomacancerstemcells,wetargeteditsexpressionusinglentivirally transduced short hairpin RNA (shRNA).
