Advanced FECD involves a thickening of the cornea from stromal edemaandchangesinDescemetmembrane.TounderstandtherelationshipbetweenFECDandcentralcornealthickness (CCT),wecharacterizedcommongeneticvariationinCOL8A2andTCF4,genespreviouslyimplicatedinCCTand/orFECD. OthergenespreviouslyassociatedwithFECD(PITX2,ZEB1,SLC4A11),andgenesonlyknowntoaffectCCT(COL5A1,FOXO1, AVGR8,ZNF469)werealsointerrogated.FECDprobands,relativesandcontrolswererecruitedfrom32clinicalsites;atotalof 532casesand204controlsweregenotypedandtestedforassociationofFECDcase/controlstatus,a7-stepFECDseverity scaleandCCT,adjustingforageandsex.AssociationofFECDgradewithTCF4washighlysignificant(OR
