DNAPolymorphismsandBiocontrolofBacillus AntagonistictoCitrusBacterialCankerwithIndicationof theInterferenceofPhyllosphereBiofilms Tzu-PiHuang1*,DeanDer-SyhTzeng1,AmyC.L.Wong2,Chun-HanChen1,Kuan-MinLu1,Ya-HueiLee1, Wen-DiHuang1,Bing-FangHwang3,Kuo-ChingTzeng1 1Department of Plant Pathology, National Chung-Hsing University, Taichung, Taiwan, 2Department of Bacteriology, University of Wisconsin-Madison, Madison, Wisconsin,UnitedStatesofAmerica,3DepartmentofOccupationalSafetyandHealth,ChinaMedicalUniversity,Taichung,Taiwan Abstract Citrus bacterial canker caused by Xanthomonas axonopodis pv. citri is a devastating disease resulting in significant crop lossesinvariouscitruscultivarsworldwide.Abiocontrolagenthasnotbeenrecommendedforthisdisease.Toexplorethe potentialofbacillinativetoTaiwantocontrolthisdisease,Bacillusspecieswithabroadspectrumofantagonisticactivity againstvariousphytopathogenswereisolatedfromplantpottingmixes,organiccompostandtherhizospheresoil.Seven strainsTKS1-1,OF3-16,SP4-17,HSP1,WG6-14,TLB7-7,andWP8-12showingsuperiorantagonisticactivitywerechosenfor biopesticidedevelopment.Thegeneticidentitybasedon16SrDNAsequencesindicatedthatallsevennativestrainswere closerelativesoftheB.subtilisgroupandappearedtobediscretefromtheB.cereusgroup.DNApolymorphismsinstrains WG6-14,SP4-17,TKS1-1,andWP8-12,asrevealedbyrepetitivesequence-basedPCRwiththeBOXA1Rprimersweresimilar toeachother,butdifferentfromthoseoftherespectiveBacillustypestrains.However,moleculartypingofthestrainsusing either tDNA-intergenic spacer regions or 16S–23S intergenic transcribed spacer regions was unable to differentiate the strainsatthespecieslevel.StrainsTKS1-1andWG6-14attenuatedsymptomdevelopmentofcitrusbacterialcanker,which wasfoundtobecorrelatedwithareductionincolonizationandbiofilmformationbyX.axonopodispv.citrionleafsurfaces. The application of a Bacillus strain TKS1-1 endospore formulation to the leaf surfaces of citrus reduced the incidence of citrusbacterialcankerandcouldpreventdevelopment
