X-ray phasing was initiated by molecular replacement, using density from cryo-electron microscopy(cryo-EM).Phaseswereimprovedbydensitymodification,includingconcentric24-and48-foldrotational symmetryaveraging. Fromthis, thecontinuous cryo-EMelectron densityseparated intodomain-like blocks.A draft atomicmodelofcpMVPwasfittothisimproveddensityfrom15domainmodels.Threedomainswereadaptedfroma nuclear magnetic resonance substructure. Nine domain models originated in ab initio tertiary structure prediction. ThreeC-terminaldomainswerebuiltbyfittingpoly-alaninetotheelectrondensity.Locationsofloopsinthismodel providesitestotestvaultfunctionsandtoexploitvaultsasnanocapsules.
